亚麻籽中开环异落叶松树脂酚二葡萄糖苷的测定——高效液相色谱-二极管阵列检测电喷雾质谱法

摘要

The crude glucosinolates (GSs) were extracted from bolting stems of Chinese kale (Brassica alboglabra) and purified by a column packed with DEAE Sephadex A-25. The desulpho-glucosinolates were obtained after desulfurizing of GSs by sulfatase. It was found that desulpho-glucosinolate compounds were completely separated by HPLC with C18 column of 250 mm × 4.6 mm (Prontosil ODS2, grain diameter of 5 μm) and the mobile phase of the acetonitrile/water system in contrast to the incomplete separating results with C18 column of 150 mm × 4.6 mm and/or the mobile phase of the methanol/water system. Finally, 11 GS compounds in Chinese kale bolting stems were separated and identified by both electrospray mass spectrometry and ultraviolet spectroscopy, including seven kinds of aliphatic GSs and four kinds of indolyl GSs. Among them, 4-methylthiobutyl GS was detected in Chinese kale for the first time. Aliphatic GSs were the dominant type of GSs in Chinese kale, accounting for 91.6% of the total GSs content, whereas, the proportion of indolyl GSs was only 8.4%. The main components of GSs were 3-butenyl GS and 4-methylsulfinylbutyl GS, accounting for 47.9% and 35.5% of the total GSs content, respectively. While the lowest content of GS was 4-hydroxy-3-indolylmethyl GS, accounting for 0.2% of the total GSs content.
