Screening of Combination of Essential Oil Components with Synergistic Inhibitory Effect on Quorum Sensing with Hexanal(与己醛具有群体感应协同抑制效应的精油成分组合筛选研究)

In order to reduce the use of essential oil as a preservative, the combinations of essential oil components were chosen and their synergistic inhibition on the quorum sensing of Erwinia carotovora was studied. Based on the minimum inhibitory concentration (MIC) and fractional inhibitory concentration (FIC) of Erwinia carotovora, four combinations of essential oil components exhibiting synergistic antibacterial effect with hexanal were selected. Subsequently, Chromobacterium violaceum 026 (CV026) and Erwinia carotovora were adopted to screen the essential oil combinations with synergistic inhibitory effect on quorum sensing was presented. Furthermore, Q value was used to verify that only the combination of hexanal and geraniol showed a better synergistic quorum sensing inhibitory effect. Similarly, the effective concentration of this combination was about 1/8 to 1/16 of that in either hexanal or geraniol. Meanwhile, this combination showed a reasonably high inhibitory effect on the quorum sensing-related indicators of CV026 and Erwinia carotovora. Among them, the inhibition rates of violacein production, swimming, swarming, extracellular polysaccharide production, and biofilm formation were 52.91%, 58.19%, 51.49%, 65.03%, and 69.84%, respectively. The combination of hexanal and geraniol showed an obvious destructive effect on biofilm formation of Erwinia carotovora observed by scanning electron microscope and confocal laser scanning microscope. The results showed that this combination which had synergistic inhibitory effect on the quorum sensing of spoilage bacteria could effectively inhibit the spoilage characteristics of spoilage bacteria while reducing the actual use of essential oil components.(为了减少精油作为抗菌剂的实际使用量,研究了对胡萝卜软腐欧氏杆菌(蔬菜腐败菌)的群体感应具有协同抑制作用的精油成分组合。基于胡萝卜软腐欧氏杆菌的最小抑菌浓度和分级抑菌浓度,从9种精油成分中筛选出与己醛具有协同抑菌作用的4个精油成分组合,利用紫色杆菌CV026和胡萝卜软腐欧氏杆菌筛选其中对二者群体感应具有协同抑制效应的精油成分组合,进一步利用能够判别两种物质是否具有协同作用的金氏公式验证,发现己醛只有与香叶醇组合才具有较好的群体感应协同抑制效果,而且有效浓度仅为己醛和香叶醇单体最小抑菌浓度的1/8和1/16。该组合对紫色杆菌CV026和胡萝卜软
