Alteration of p53 suppression gene in gastric mucosa and carcinogenesis

AIM To study the alteration of p53 in a variety of gastric mucosal lesions. METHODS Immunohistochemical staining was used to examine the overexpression of p53 protein in 154 cases of human gastric mucosal lesions from endoscopic biopsies. We also used PCR-SSCP analysis and DNA sequence to detect p53 exon 5 to 8 mutation. RESULTS Positive nuclear staining for p53 protein was 33 3%(10/30 cases) in gastric carcinoma except non-malignant mucosal lesions, and p53 mutation of exon 5 to 8 was determined in 6 7% (1/15 cases) of intestinal metaplasia, 20% (3/15 cases) of dysplasia, and 54 5% (6/11 cases) of gastric carcinoma. We confirmed p53 misense mutation (G→T transversion) and delete C of exon 5 in gastric carcinoma and misense mutation (G→C transversion) of exon 6 in dysplasia by using DNA sequence analysis. CONCLUSION Alteration of p53 suppression gene may be closely associated with the malignant transformation of gastric mucosa.
