A codominant SCAR marker linked to SMV resistance gene Rsa

The conversion of RAPD (randomly amplified polymorphic DNAs markers to sequence characterized amplified regions (SCAR) counterparts has been proven to be an effective method to obtain more useful and reliable markers. Nine hundred random 10-mer primers were screened using bulked segregant analysis (BSA). Two dominant RAPD markers, OPAS-061800 and OPW-05660, were identified to be linked to Rsa, a single dominant gene resistant to a soybean mosaic virus (SMV) strain Sa. The marker OPW-05660 close to Rsa was cloned and sequenced Two ends of the sequence were used to design a pair of specific oligonucleotide primers. The pair of primers generated polymorphic fragments between resistant parent and susceptible parent, indicating the dominant RAPD marker was successfully converted to a codominant SCAR marker SCW-05 . The genetic distance between SCW-05 and Rsa is 7 .7 cM. The genetic diversity detected by SCW-05 was consistent with resistance or susceptibility in 14 soybean cultivars.
