THE CONSTRUCTION OF A UNIVERSAL VECTOR pGJP-5 FOR GENE EXPRESSION IN VACCINIA VIRUS
摘要
We have construeted a universal vector pGJP-5 for gene expression in vaccinia virus. A low side-effect Chinese strain, Guang-9, was utilized as a starting virus. The thymidine kinase (TK) gene of Guang-9 in the vector provided not only the homologous sequences for in rive recombination with the vaccinia virus genome, but a so the selective marker for the virus recombinants. The promoter of the gene coding for vaccinia viral 7.5 K protein (P_(7.5)) was inserted into the TK structure gene. On the downstream side of P_(7.5), several restriction sites, BamHI, Sinai, SaeI, EcoRI, were arranged for the insertion of the foreign genes. The inserted foreign gene can be expressed efficiently under the control of the promoter P_(7.5).
