纳米微囊-VEGF复合体对创伤组织中MMP-9及TIMP-1mRNA表达的影响
摘要
Objective: To study the effects of complex of nanosphere-VECF on expression of MMP-9 and TIMP-1mRNA, and investigate its mechanisms of improving wounded tissue repairing. Methods: We excerted complex of nanosphere-VECF on wounded rabit ears, samples were divided into chronic wound + VEGF-transgenic group (CW+VEGF), chronic wound group (CW) and nonischemia wound group (NW), using RT-PCR was used to examine expression of MMP-9 and TIMP-1mRNA on 1d, 3d, 7d, 10d, 14d post operation. Results: ① The expression of MMP-9mRNA of rabbits in CW was greater than in NW at the same time from 3d to 14d post operation, with statistical significance (P<0.05). ② The expression of TIMP-1mRNA in NW and CW+VEGF was both greater than in CW at the same time from 3d to 14d post operation, with statistical significance (P<0.05). Conclusion: High expression of MMP-9 and low expression of TIMP-1 might be important mechanisms of chronic raw surface healing exogenous VEGF gene transfection might promote wound healing through upregulating the expression of TIMP-1.
