高热惊厥发育期大鼠脑组织血红素氧合酶-1 mRNA表达及锌原卟啉对其影响

Objective To explore the effect of Znic Protoporphyrin (ZnPP) and its mechanism on the hemeoxygenase -1 (HO-1) mRNA expression in brain tissue of developing rat with febrile convulsion (FC). Methods Sixty-five21-day Wistar rats were randomly divided into 4 groups: control group (n=14), none -FC group (0=16), FC group (n=20) and ZnPP treated groups (n=15). FC in rats was induced 10 times, once every 2 days in a bath of warm water. ZnPP treated group were received ZnPP 45 µmol/kg before FC models. The expressions of HO -1 mRNA were detected in hippocampal CA1, CA3 and dentate gyros with the technique of hybridization in situ. Results The expressions of HO -1 mRNA significantly increased in hippocampal CA1, CA3 region and dentate gyros in PC group compared with those in control group ,none PC group and ZnPP treated group (P(subscript a)<0.01). And the HO -1 expression in ZnPP -treated group was significantly higher than that of none FC group and control group (P(subscript a)<0.01). Conclusions Recurrent FC in rats can cause the increase of HO -1 mRNA expression; ZnPP probably plays a role in protecting the brain from PC damage by down -regulating the HO -1 mRNA expression.
